For the first time in his presidency, Barack Obama can call himself a job creator.

There are now more U.S. jobs than when he took office. Not by much, but they've moved out of negative territory, according to a Labor Department revision.

It probably won't help Obama much in his race for re-election. The economy is still a long way from firing on all cylinders.

But the revision ? showing employers added 386,000 more jobs than had previously been estimated in the 12 months ended in March ? was clearly good news for the White House as Obama heads into a crucial week.

The focus of next Wednesday's first presidential debate will be the economy. On Friday the September employment report comes out.

Economists don't expect much change in the jobless figures from August's 8.1 percent unemployment rate.

Still, "we are making progress," said Alan Krueger, chairman of Obama's Council of Economic Advisers. The White House can now claim a net gain of about 100,000 jobs under Obama.

Still, unemployment has been stuck above 8 percent since early in Obama's term, budget deficits have topped $1 trillion for four consecutive years and economic growth has slowed to a trickle, expanding at a scant 1.3 percent annual rate in the April-June quarter.

Republican challenger Mitt Romney portrays Obama as a failed job creator. Romney says he would create 12 million jobs if he's elected president.

Polls have suggested a recent shift in public opinion, with some for the first time showing Obama ahead on handling the economy.

Romney campaigned in Pennsylvania Friday. "My priority is job creation and growing incomes," Romney told donors in Philadelphia "My priority is not trying to punish people who have been successful." He also attended a rally in Wayne, Pa.

Obama had three fundraisers, all in Washington, D.C.

(AP Photo/Gene J. Puskar, File)

NEW YORK (AP) ? A key witness against convicted child molester and former Penn State assistant football coach Jerry Sandusky, known in court papers as Victim 1, has a book deal and will soon reveal his identity, a publisher announced Thursday.

Ballantine Bantam Dell said that "Silent No More: Victim (hash)1's Fight for Justice Against Jerry Sandusky," is coming out Oct. 23. The memoir will be co-written by the victim's mother and psychologist and "will share how he survived years of shame and secrecy, harassment and accusation, before reporting Sandusky's actions to the authorities, and will offer a hopeful and inspiring message for victims of abuse," Ballantine announced.

Victim 1, now 18, will reveal his identity on the day of the book's release in an interview with ABC's Chris Cuomo.

Financial terms for the book were not disclosed. But Ballantine, an imprint of Random House Inc., plans a donation to a charity for victims of child abuse.

Victim 1 first alerted authorities in 2008 and helped launch the investigation leading to Sandusky's conviction in June on 45 counts of child sexual abuse. Prosecutors said some of the assaults took place on the Penn State campus. Sandusky is scheduled to be sentenced next month and is likely to receive a sentence that will keep him in prison for life.

Victim 1 testified for the prosecution that Sandusky approached him through a summer camp for youth sponsored by The Second Mile, a charity for at-risk youth that the former coach had founded. Their initial contacts consisted of football games and swimming, and Sandusky would attend the boy's wrestling tournaments.

Physical contact began with a hand on his leg in the car, Victim 1 said, and he began spending nights at the Sandusky home in State College, about 30 miles from his own home in Lock Haven.

Kissing and back rubbing during those overnight visits, when he was in his early teens, progressed to oral sex, he testified, sobbing. He said Sandusky eventually told him: "It's your turn."

"I don't how to explain it, I froze," he said. "My mind is telling me to move but I couldn't do it, I couldn't move."

He told jurors his own behavior worsened, going from a well-behaved child to fights with relatives and bed wetting. His grades deteriorated. After Victim 1 began trying to avoid Sandusky, he asked his mother about websites for child molesters, "to see if Jerry was on there," he testified.

His mother set up a meeting with school guidance counselors, a process that led to the opening of a police investigation and produced criminal charges against Sandusky in November. Messages seeking comment left Thursday for Victim 1's civil lawyers, Slade McLaughlin and Michael Boni, were not immediately returned.

Researchers determine how inflammatory cells function, setting stage for future remedies

Researchers determine how inflammatory cells function, setting stage for future remedies

Thursday, September 27, 2012

A research team led by investigators at New York University and NYU School of Medicine has determined how cells that cause inflammatory ailments, such as Crohn's disease, multiple sclerosis, and arthritis, differentiate from stem cells and ultimately affect the clinical outcome of these diseases.

"We've found that hundreds of new genes are involved in the function and development of these cells," said co-author Richard Bonneau, an associate professor at New York University's Center for Genomics and Systems Biology and the Courant Institute of Mathematical Sciences. "This expansion in our understanding can be used as a framework for designing new therapies to combat a range of ailments where the immune system attacks self."

These cells, called T-cells by immunologists, play a role in fighting off infection, but can also induce inflammation and other processes that damage tissues and contribute to several common inflammatory diseases. T-cells are also key cell types in new immune-cell based therapies for fighting cancer. There are many types of T-cells, and how they differentiate from stem cells in the human body lies at the center of understanding several diseases.

"We have been striving for several years to understand what makes inflammatory T-lymphocytes special," said lead investigator Dan Littman, MD, PhD, the Helen L. and Martin S. Kimmel Professor of Molecular Immunology and a professor of pathology, microbiology and molecular pathogenesis at NYU School of Medicine and a faculty member of the Skirball Institute on Biomolecular Medicine. "They can protect us from microbes, but they also have the potential to cause autoimmune disease.

"We were fortunate to be able to bring together a team of immunologists, computational biologists, and genomics experts to begin to solve this puzzle. Whereas before we only knew of a handful of genes that influence the function of these cells, we now know of hundreds of new ones that can serve as a resource for further studies by us and other laboratories. Our hope is that some of these new molecules will be the Achilles heel that we can target to treat these diseases."

The findings, which are reported in the latest issue of the journal Cell, lay the groundwork for understanding how these cells regulate their genomes through a regulatory network that connects many environmental stimuli to a large number of genes and their interactions. This large network model is essentially the brain that T-cell precursors, or stem cells, use to decide what they want to be when they grow up. Specifically, a network model can be used simulate what inhibiting a gene with a drug would do to different T-cells and, in this way, aid the development of new therapeutic measures to address these afflictions.

The study focused on T-helper 17 cells (Th17) and how they regulate the synthesis of gene products from thousands of regions of the chromosome. Th17 cells have previously been implicated in inflammatory diseases. Other studies have also identified hundreds of genes that roles in pro-inflammatory diseases. This new study places these implicated genes on a timeline of cellular development and ultimately puts them together in an integrated model of how genes interact.

To explore the inner workings of these cells, the researchers used a systems biology approach, which focuses measuring multiple biomolecules and capturing multiple interactions within an organism to understand how it functions. For example, each of the 450 data sets integrated in this study contained measurements of gene expression, chromatin structure, or gene-chromosome interaction that spanned millions of locations along the genome. This holistic method offers a broader understanding of interconnected molecular phenomena essential to running life's program?a process similar to studying an entire automobile while it functions rather than separately studying the headlights, brakes, or steering column.

To extract meaningful results from this very large data set, the researchers employed statistical techniques to uncover the network model from the large amount of data. To verify the accuracy of the computer modeling, further laboratory experiments were conducted using mice. Although the study was carried out in mice, the researchers found that their work could offer explanations as to why a large number of genes are associated with several human inflammatory diseases. The researchers' computer models identified candidate genes that influence the expression of more than 2,000 genes and play a significant role in the regulation of Th17 cells. They found that the core of this network was significantly enriched for the genes that human geneticists have found to be associated with pro-inflammatory disease. These genes serve as cogs in the regulation of expression or suppression of genes in Th17 cells and are potential leads for developing new therapeutic approaches to modulating inflammation.

On video, dolphin gives birth to calf in Hawaii

HONOLULU (AP) ? A 12-year-old dolphin at a Hawaii resort has given birth to a female calf that seems to instantly recognize her mother in a video of the birth posted online.

Footage of last week's birth on the Big Island shows the baby dolphin's tail moments before she emerges from her mother. Once she is born, she shoots up to the water's surface to take her first breath, then quickly swims alongside her mother.

The birth occurred in a manmade lagoon at Dolphin Quest Hawaii at the Hilton Waikoloa Village, where visitors can touch and swim with the marine mammals.

Resort officials will monitor the baby around the clock for now, as its first 30 days of life are its most critical in terms of survival, said Julie Rocho-Levine, manager of marine animals for Dolphin Quest. Trainers will closely note when the baby nurses, among other things, she told The Associated Press on Monday.

Officials say it's the first calf for the mother, Keo.

"I'm a mom myself, so I feel like I was able to appreciate her just calm, relaxed nature throughout the whole entire situation," said Rocho-Levine, who was there for the birth.

"It seemed as though she (Keo) was seeking out that human companionship and finding comfort in the people she knows and spends her days with," she said.

Keo was calm enough to allow veterinarians to perform an ultrasound during labor.

Dolphin Quest officials plan to wait to name the baby until after its first month of life.

The rate of survival for babies of first-time mother dolphins in the wild is about 50 percent, Rocho-Levine said. But that rate is much higher for dolphins born with access to top-notch care from humans, she said.

Marilee Menard, executive director of the Alliance of Marine Mammal Parks and Aquariums, said 70 percent of the dolphins in accredited facilities in North America were born in a zoo or aquarium.

Air Force launching secretive X-37B space plane

The U.S. military's hush-hush robotic X-37B space plane is slated to blast off again next month, Air Force officials say. The mission will test the robotic spacecraft's reusability and may eventually land on the Florida runway once used for NASA space shuttles.

The X-37B space plane 's next mission ? called Orbital Test Vehicle-3, or OTV-3, because it is the program's third-ever spaceflight ? is scheduled to launch aboard an Atlas 5 rocket from Florida's Cape Canaveral Air Force Station (CCAFS) sometime in October.

"Preparations for launch at Cape Canaveral have begun," said Major Tracy Bunko at the Pentagon?s Air Force press desk. "We are on track to launch OTV-3 next month; however, the exact date remains subject to change based on range conditions, weather, etc."

A mysterious mission
As with the X-37B program's two previous spaceflights ? OTV-1 and OTV-2 ? OTV-3's payload and mission details are classified. But the focus remains on testing vehicle capabilities and proving the utility and cost-effectiveness of a reusable spacecraft, Bunko told SPACE.com. [ Photos: The X-37B Space Plane ]

Bunko said in an earlier communiqu? that this third flight will use the same X-37B spacecraft that flew the first test flight, the OTV-1 mission, back in 2010.

That maiden voyage of the miniature space plane lasted 225 days. It launched into orbit on April 22, 2010, and then landed on Dec. 3 of that year, zooming in on autopilot over the Pacific Ocean and gliding down onto a specially prepared runway at Vandenberg Air Force Base in California.

  1. Space news from NBCNews.com

    1. Beyond-the-moon base stirs up buzz

Science editor Alan Boyle's blog: A concept that calls for building a deep-space outpost beyond the moon's orbit has stirred up some positive buzz from space pioneers ? including Apollo 11 moonwalker Buzz Aldrin.

    2. Dazzling meteor fireball lights up UK sky

    4. Romney reveals space exploration plans

A different X-37B vehicle made a similar Vandenberg touchdown this past June 16, having stayed in orbit for 469 days on its OTV-2 mission.

The X-37B program is being run by the U.S. Air Force Rapid Capabilities Office. The two space planes ? which are 29 feet (8.8 meters) long and 15 feet (4.5 m) wide, with a payload bay about the size of a pickup truck bed ? were built by Boeing Government Space Systems.

While they're sparing with details about the X-37B program, Air Force officials say the vehicles enable them to test out how new technologies perform in space.

?One of the most promising aspects of the X-37B is it enables us to examine a payload system or technology in the environment in which it will perform its mission and inspect them when we bring them back to Earth,? Bunko said. ?Returning an experiment via the X-37B OTV enables detailed inspection and significantly better learning than can be achieved by remote telemetry alone.?

A new landing site?
While both previous X-37B missions touched down at Vandenberg, the Air Force is considering landing future flights at NASA's Kennedy Space Center in Florida, next door to the Cape Canaveral launch site.

In fact, the Air Force is currently conducting taxi and braking tests as part of an ongoing appraisal. The prospect of a returning the robotic space plane to the KSC landing strip ? which was used by NASA's now-retired space shuttle fleet ? is seen as a cost-saving measure.

"We are also considering consolidating landing, refurbishment and launch operations at KSC or CCAFS in an effort to save money," Bunko said.

?We are seeking to leverage previous space shuttle investments and are investigating the possibility of using the former shuttle infrastructure for X-37B OTV landing operations," Bunko added. "Those investigations are in an early state, and any specifics will not be known for some time, but could potentially be used as early as for the landing of OTV-3."

Schools Jump onto Free Online Course Service but Hedge on Certification

Coursera's on a growing streak. The number of new institutions of higher education to sign onto the free online course site has more than doubled, bringing the total count of colleges and universities participating to 33 from 16. Among the new schools participating: Berklee College of Music, Columbia University, Ohio State, Vanderbilt, and Wesleyan.

Each participating institution contracts with the company to provide classes taught by members of their faculty. With the new additions, Coursera now hosts more than 200 courses from 33 domestic and international universities and reaches 1.3 million students, or "Courserians," as they're called, around the world.

Students can sign up for free to gain access to online video lectures and interactive activities, such as discussion forums. The initiative has also sparked creation of in-person meetups in 600 cities, according to the company. The classes, which also include required reading, assignments, and quizzes, last a set number of weeks and in many cases earn a certificate at the end for those who successfully complete the requirements.

One new participant is the University of California Irvine, which has added seven courses and is already involved in a Coursera-like initiative run by the OpenCourseWare Consortium, but without the interactive components that Coursera provides. UCI also offers educational materials on iTunes U, YouTube, Connexions, California State University-managed MERLOT, and other repositories.

So making the decision to join Coursera wasn't a big leap, according to Gary Matkin, dean of Continuing Education, Distance Learning, and Summer Session, and Larry Cooperman, director of UCI's OpenCourseWare. "It's another channel, another way of getting our stuff out there," notes Matkin.

Good Deal: get Logitech's Ultrathin iPad keyboard cover for only $39.99

Logitech's Ultrathin Keyboard Cover for iPad was one of the best options we found in our exhaustive roundup, and for the next few hours you can bring home a refurbished model for just $39.99 at 1saleaday. The Bluetooth keyboard (normally $99.99) doubles as a Smart Cover replacement, with the rear aluminum finish complementing your new iPad or iPad 2 rather nicely. And though we found the keys to be a bit plasticky on account of thinness, the overall package still carries a high recommendation.

Keep in mind, however, that buying refurbished means you'll receive just 90 days of warranty coverage rather than the three years that accompany new purchases. Having said that, Logitech's hardware reputation should alleviate most concerns, and more importantly, you're saving an astonishing $60 over the regular pricing. It's a limited-time deal and minutes are ticking away, so we'd recommend getting in soon if you're on the hunt for an iPad typing companion.

Iran creates domestic Internet network: report

1 hr.

DUBAI ??Iran has connected all its government agencies to a secure domestic Internet service and plans to link ordinary Iranians up to the same network, an official was quoted as saying on Sunday, in a move to beef up cyber security.

The Islamic state tightened its cyber security after its disputed nuclear program was attacked in 2010 by the Stuxnet computer worm, which caused centrifuges to fail at the main Iranian uranium enrichment facility.

Tehran, whose nuclear program is suspected by the West of being aimed at developing a bomb, accused the United States and Israel of deploying the worm.

"In recent days, all governmental agencies and offices ... have been connected to the national information network," deputy communications and technology minister Ali Hakim-Javadi was quoted as saying on Sunday by the Mehr news agency.

The second phase of the plan would be to connect ordinary Iranians to the national network.

According to Iranian media, the domestic system would be fully implemented by March 2013 but it was not clear whether access to the global Internet would be cut once the secure Iranian system was rolled out.

Millions of websites deemed to have un-Islamic content are blocked by Iranian authorities, along with many expressing anti-government views.

Many Iranians suffered serious problems accessing email and Internet social networking sites in February, ahead of parliamentary elections.

Opposition supporters used social networking sites to organize widespread protests after the disputed 2009 re-election of President Mahmoud Ahmadinejad, which they said was rigged in his favor.

Communications and Technology Minister Reza Taqipour said last month Iran needed to develop its own network to ensure the safety of the country's information, the Fars news agency reported.

"Especially on major issues and during crises, one cannot trust this network at all," he said, referring to the global Internet.

"Control over the Internet should not be in the hands of one or two countries."

Authorities said in April a computer virus was detected inside the control systems of Kharg Island ?which handles the vast majority of Iran's crude oil exports ? but the terminal remained operational.?

Romney Solidifies Major Shift On Health Care (VIDEO) - TPMDC

Comments made by Mitt Romney on Sunday night to CBS News reveal again, in stark relief, how fully he?s abandoned the basic tenets of the health care reform law he enacted in Massachusetts.

?The guy has come completely full circle,? says Jonathan Gruber, a professor at MIT who advised Romney on the Massachusetts law and has expressed his dismay about Romney?s shift in several public fora.

In the 60 Minutes interview, Romney protested the idea that government doesn?t already provide health care to the uninsured: ?Well, we do provide care for people who don?t have insurance,? he said. ?If someone has a heart attack, they don?t sit in their apartment and die. We pick them up in an ambulance, and take them to the hospital, and give them care. And different states have different ways of providing for that care.?

The new message reflects a Republican Party consensus against using government power to help insure the tens of millions of Americans who lack coverage ? the Affordable Care Act?s key accomplishment. In 2008, President George W. Bush created an election year controversy when he boasted, ?[P]eople have access to health care in America. After all, you just go to an emergency room.? But for Romney, it?s the culmination of a years? long conservative pressure campaign, which has forced him to abandon the premise of his 2006 Massachusetts health care law.

To fully appreciate the enormity of Romney?s shift, you need go no further than Romney?s own book or a series of public statements he made, some as recently as 2010.

?After about a year of looking at data ? and not making much progress ? we had a collective epiphany of sorts, an obvious one, as important observations often are: the people in Massachusetts who didn?t have health insurance were, in fact, already receiving health care,? Romney wrote in No Apology.

Under federal law, hospitals had to stabilize and treat people who arrived at their emergency rooms with acute conditions. And our state?s hospitals were offering even more assistance than the federal government required. That meant that someone was already paying for the cost of treating people who didn?t have health insurance. If we could get our hands on that money, and therefore redirect it to help the uninsured buy insurance instead and obtain treatment in the way that the vast majority of individuals did ? before acute conditions developed ? the cost of insuring everyone in the state might not be as expensive as I had feared.

?The whole idea of Romneycare was to avoid this kind of free riding,? Gruber told TPM.

The Republican nominee made the same point while running for president in 2008. In March 2010, the year ?Obamacare? was enacted, he was still arguing that the guarantee of emergency room coverage ought to be supplemented by bringing Americans into the insurance system.

?It doesn?t make a lot of sense for us to have millions and millions of people who have no health insurance, and yet who can go to the emergency room and get entirely free care for which they have no responsibility, particularly if they?re people who have sufficient means to pay their own way,? Romney said at the time on MSNBC?s ?Morning Joe.? ?And so we said, look, we?ve got an idea. Let?s take all the money we?ve been spending to give out free care ? paying hospitals who give out free care ? and help people who can?t afford it buy insurance.?
